Cayman Chemical HDAC Fluorometric Activity Assay Kit, 96 Well
Cayman’s HDAC Fluorometric Activity Assay Kit provides a fast, fluorometric method for measuring HDAC activity without the use of radioactivity, extraction, or chromatography. In the first step, an acetylated lysine substrate is incubated with samples. Deacetylation sensitizes the substrate such that treatment with the HDAC developer in the second step releases a fluorescent product. Trichostatin A is included as a control inhibitor. The fluorophore can be easily analyzed using a fluorescence plate reader with an excitation wavelength of 340-360 nm and an emission wavelength of 440-465 nm. The assay can be used for quantifying Class I and II HDAC activity from various sources.

Cayman Chemical Neutrophil Elastase Activity Assay Kit, 192 Well
Cayman’s Neutrophil Elastase Activity Assay kit employs a specific non-fluorescent elastase substrate, (Z-Ala-Ala-Ala-Ala)2Rh110, which is selectively cleaved by elastase to yield the highly fluorescent compound R110. Fluorescence can be analyzed with an excitation wavelength of 485 nm and emission wavelength of 525 nm. Reagents needed to isolate neutrophils from whole blood are included in the kit, as is PMA, which is known to stimulate elastase release from neutrophils. The assay can be used to study compounds regulating elastase release in neutrophils. If a cell line known to express neutrophil elastase is used, a proper control using a specific neutrophil elastase inhibitor, such as ONO-6818 which is not provided, should be included to ensure specificity of the enzyme.

Mechanism of Action A cationic amphiphilic tracer (CAT) dye that rapidly partitions into cells in a similar manner as drugs that induce phospholipidosis. This tracer was created by placing titratable groups on the probe (dye), to enable labeling to be expanded into lamellar inclusion bodies of cells pretreated with weakly basic cell-permeant compounds.
